TaqMan is a PCR method that exponentially amplifies the DNA and measures real-time after each amplification cycle using fluorescent TaqMan probes.
TaqMan primers and probes are designed in such a way that they anneal within a specific DNA region of a target organism. The TaqMan probe consists of a fluorophore covalently attached to the 5'-end of the oligonucleotide probe and a quencher at the 3'-end. During PCR amplification the fluorophore will be cleaved away and is decoupled from the quencher. This leads to a strong increase in fluorescence intensity, which is proportional to the specific target DNA amplification. This fluorescence is directly measured and quantified in the real-time thermocycler.
